In the present study chitosan – mango leaf extract microspheres were used as the potential carrier for the release studies of the antidiabetic drug metformin hydrochloride. A series of microspheres were prepared with varying compositions of chitosan and mango leaf extract, in sodium tripolyphosphate (TPP) by the ionotropic gelation method. Spectroscopic studies and surface morphology of the prepared microspheres were conducted. The swelling properties, percentage yield, drug loading, encapsulation efficiency, drug release, cytotoxicity and antidiabetic activity of the prepared microspheres were analysed. The swelling capacity of the metformin loaded microspheres significantly depends on the ratio of chitosan and mango leaf extract in the microsphere samples. The drug release studies disclosed that the release rate is directly proportional to the swelling index of the drug carrier. The nontoxic nature of the drug loaded chitosan mango leaf extract microspheres was confirmed by MTT assay using L929 (mouse fibroblast) cells. The antidiabetic activity of metformin loaded chitosan – mango leaf extract microspheres was done by following α-glucosidase inhibitory activity.
